Mycoplasma testing of ATCC VR-1477 by direct method in broth and agar showed positive results.
FBS used to culture Chlamydia must be prescreened to verify that the serum does not contain antibodies to Chlamydia or other factors that would interfere with growth.
This strain is a tissue culture-adapted version of strain TW-3 (VR-578) which is grown in embryonated chicken eggs.
Before infecting host cells, add 1-mm glass beads and vortex stock material for 45 seconds. Aspirate fluid. Infect shell vial monolayer with 0.2 mL of material.
Centrifuge at 800 x G for 1 hour. Feed with 1.0 mL EMEM (HEPES buffered), 10% FBS (screened for chlamydia antibodies).

Next-generation sequencing (NGS) at ATCC on the McCoy cell line (ATCC® CRL-1696™) used as the host has shown the presence of Mus Musculus mobilized endogenous polytropic provirus and Murine leukemia virus.

Growth Conditions
Temperature: 37°C
Recommendations for Infection: Add glass beads and vortex preparation to disrupt cells. Infect monolayer with disrupted material. Centrifuge at 3000 x rpm (750 x g) for 1 hour. Feed with fresh growth medium containing FBS prescreened for Chlamydia antibodies and 1-2mg/mL cycloheximide. Incubate at 37°C for 48 to 72 hours, a 5% CO2 in air atmosphere is recommended.
