Restriction digests of the clone give the following sizes (kb): BamHI--5.6; EcoRI/PstI--3.6, 2.1; HindIII--5.4, 0.23; PstI--5.6; HincII--5.6. This probe was derived from a 7.5 genomic insert isolated from the chromosome X library of Davies et al. This probe contains a unique sequence (repeat free). This probe detects a fragment mapping to Yq11 which is 80% homologous and is slightly smaller than 3.4 kb. Detection of the Y fragment can be prevented by washing at 65C in 0.5X SSC. The fragment can also be isolated by double digestion with EcoRI and HhaI. The vector is in fragments less than 400 bp. Enzyme(s) not detecting polymorphism: BamHI, BanI, BanII, BclI, BglI, BglII, BstNI, DraI, EcoRI, EcoRV, MspI, PvuII, SacI, ScaIl, StuI, TaqI, Tth111I, XbaI. |
